[体外研究]

萝卜硫素以剂量依赖性方式诱导细胞周期停滞,随后细胞死亡。该萝卜硫素诱导的细胞周期停滞与细胞周期蛋白A和B1的表达增加相关。 Sulforaphane通过凋亡过程诱导细胞死亡。 Sulforaphane抑制生长的重新开始并减少静息结肠癌细胞(HT29)中的细胞活力,并且对分化的CaCo2细胞具有较低的毒性[1]。用萝卜硫素预处理H9c2大鼠成肌细胞减少了凋亡细胞数量和促凋亡蛋白(Bax,caspase-3和细胞色素c)的表达,以及阿霉素诱导的线粒体膜电位的增加。此外,萝卜硫素增加血红素加氧酶-1的mRNA和蛋白质表达,从而降低线粒体中由阿霉素诱导的活性氧物质(ROS,使用MitoSOX Red试剂测量)[2]。

[体内研究]

在用单剂量的9,10-二甲基-1,2-苯并蒽处理的Sprague-Dawley大鼠中,萝卜硫素可以阻断乳腺肿瘤的形成。施用萝卜硫素降低了雌性Sprague-Dawley大鼠中单剂量DMBA诱发的乳腺肿瘤的发生率,多样性和重量,并延缓了其发展[3]。

[细胞实验]

将HT29细胞以低密度（5×10 4细胞/ mL）接种在含有5％FCS的标准培养基中的直径为35或120mm的Primaria培养皿中。接种后一天，更换培养基，用萝卜硫素（0-30μM）处理HT29细胞。向对照细胞中加入等量的溶剂（乙醇）（0.2％终浓度）。使用MTT测定评估药物对细胞活力的影响[1]。

[动物实验]

大鼠：在47,48,49,50和51天时，每只动物通过管饲法接受0.5mL单独的Emulphor EL-620或指定剂量（每天75,100或150μM）的萝卜硫素或化合物2， 3或4在0.5毫升Emulphor EL-620中。在给药载体或保护剂后第5天，3小时，所有大鼠也接受溶解在1.0mL芝麻油中的8.0mg DMBA的胃内滴注。选择该剂量的DMBA以产生显着的肿瘤发生率，但不高到足以淹没潜在的化学保护作用。每周一次检查动物的可触知肿瘤的外观和位置。在202天时，即致癌物施用后152天，用乙醚对所有动物实施安乐死并称重。通过解剖将肿瘤与脂肪和结缔组织分离，称重并固定在缓冲的10％福尔马林中。通过检查染色切片，在显微镜下鉴定所有肿瘤[3]。
