[体外研究]

L-亮氨酸(10mM)治疗会损害内分泌祖细胞的发育[1]。在E13.5大鼠胰腺外植体中,在没有添加L-亮氨酸的情况下,培养1天后Ngn3 mRNA水平增加,在第3天达到峰值,然后降低。当加入L-亮氨酸时,在Ngn3 mRNA水平中观察到显着降低。 Ngn3 mRNA水平的降低与Ngn3表达细胞数量的减少相平行(4728±408对959±28; P <0.01)。最后,L-亮氨酸还对三种基因的mRNA水平产生剂量依赖性抑制作用,即Ngn3,其靶标Insm1和胰岛素[1]。亮氨酸通过增强mTORC1活化刺激新生猪骨骼肌中的蛋白质合成。 L-亮氨酸增加细胞内HIF-1α水平并激活HIF-1α信号传导途径,并且这两种作用由mTOR信号传导途径介导。该过程导致Ngn3抑制,并因此降低β细胞分化[1]。
