[体内研究]

发现物质P(7-11)作为针对SP诱导的反应的非常有效的拮抗剂,并且在SP注射后在黑质中局部形成。建议物质P(7-11)是SP作用的内源调节剂[1]。注射低剂量的P物质(7-11)(与SP或SP(5-11)同时为1.0-4.0 pM(0.1 nM),可显着减少SP或SP(5- 11)引起的厌恶行为。这些结果表明内源性形成的SP(1-7)可以调节脊髓中SP或SP(5-11)的作用[2]。

[动物实验]

将体重为250-300g的大鼠[1] Sprague-Dawley雄性大鼠用氟烷麻醉并置于立体定位框架中。 SNR中插入了具有约0.15mm的穿透尖端直径的圆锥形注射套管。将盐水（0.2μL），SP（0.007-0.7nM）或P物质（7-11）（0.01-1nM）注射到左侧黑质网状物（SNR）中，并将大鼠置于旋转计中。在1分钟的时间内以0.2μL的总体积注入物质。在注射后1小时通过断头处死一组动物，立即取出它们的脑并从左右纹状体，苍白球（GP）和黑质（SN）中取出组织样品。测定样品的SP和SP（1-7）[1]。小鼠[2]在厌食反应的整个期间和在20分钟时，在雄性小鼠（STD菌株，23-28g）中测量后肢搔抓，咬，前爪和后爪舔的往复运动的累积响应时间（s）。最大值。测试物质P（7-11）抑制由鞘内注射SP或SP（5-11）（0.1nM /小鼠）产生的厌恶反应的能力。然后将物质P（7-11）（1,2,4μM）与SP或SP（5-11）一起给药[2]。
