用途： Trimethoprim is a bacteriostatic antibiotic used mainly in the prophylaxis and treatment of urinary tract infections.Target: DHFRTrimethoprim (TMP), an inhibitor of dihydrofolate reductase, decreases the level of tetrahydrofolate supplying one-carbon units for biosynthesis of nucleotides, proteins, and panthotenate. TMP caused induction of DnaK, DnaJ, GroEL, ClpB, and IbpA/B Hsps. Among these Hsps, IbpA/B were most efficiently induced by TMP and coaggregated with the insoluble proteins [1]. Trimethoprim binds to dihydrofolate reductase and inhibits the reduction of dihydrofolic acid (DHF) to tetrahydrofolic acid (THF). THF is an essential precursor in the thymidine synthesis pathway and interference with this pathway inhibits bacterial DNA synthesis. Trimethoprim's affinity for bacterial dihydrofolate reductase is several thousand times greater than its affinity for human dihydrofolate reductase. Sulfamethoxazole inhibits dihydropteroate synthetase, an enzyme involved further upstream in the same pathway. Trimethoprim and sulfamethoxazole are commonly used in combination due to their synergistic effects. This drug combination also reduces the development of resistance that is seen when either drug is used alone [2]. 作用： 用途一：作为抗菌增效药，也可以治疗家禽细菌感染和球虫病。 用途二：新型口服广谱抗菌药物。抗菌谱与磺胺药物相似而效力较强，对多种革兰氏阳性和阴性细菌有效。由于细菌对本品将易产生耐药性，故不宜单独作为抗菌药使用。甲氧苄啶与磺胺药合用可使抗菌作用增强数倍至数十倍。该品主要作磺安类药的增效药。一般按1：5的比例使用。也可作为兽药，治疗禽率大肠肝菌引起的败血症，鸡白痢，禽伤寒，霍乱，呼吸系统继发性细菌感染等。还可用于球虫病的治疗。 用途三：抗菌增效药，单独用于呼吸道感染、泌尿道感染、肠道感染等病症 物理化学性质： CAS号：738-70-5 常用中文名：甲氧苄氨嘧啶 常用英文名：Trimethoprim 分子式：C14H18N4O3 分子量：290.318 密度：1.3±0.1 g/cm3 沸点：405.2±55.0 °C at 760 mmHg 熔点：199-203 °C 闪点：198.8±31.5 °C 储存条件：2-8°C |
