Use of AMXT-1501AMXT-1501 (AMXT 1501, AMXT1501) is a novel potent polyamine transport inhibitor, synergistically reduces cell viability in NB cells in combination of with DFMO; completely blocked radiolabeled spermidine uptake in multiple NB cell lines, whereas the combination of AMXT 1501 and DFMO was synergistic in reducing colony formation in BE(2)-C and KELLY cells; The combination treatment resulted in hypophosphorylation of retinoblastoma protein (Rb), suggesting growth inhibition via G1 cell cycle arrest in NB cells; inhibits polyamine synthesis and uptake, reduces tumor progression and prolongs survival in mouse models of neuroblastoma combined with DFMO.
