甘氨酰-脯氨酰-谷氨酸用途

Gly-Pro-Glu是一种神经活性肽,对乙酰胆碱释放具有有效作用。Gly-Pro-Glu是胰岛素样生长因子-I的N-端三肽。Gly-Pro-Glu抑制谷氨酸与N-甲基-D-天冬氨酸(NMDA)受体结合,IC50值为14.7μM。Gly-Pro-Glu可用于神经保护研究[1][2]。

[体外研究]

Gly-Pro-Glu(0-100μM)增强乙酰胆碱和多巴胺的钾诱发释放,即使在0.1 nM的浓度下也能增加K+诱发的乙酰胆碱释放,并显著增强诱发的多巴胺释放[1]。Gly-Pro-Glu(1-1000μM)显示出对L-[3H]谷氨酸结合的抑制作用,IC50值为14.7μM[1]。

[体内研究]

Gly-Pro-Glu(300 mg；每天i.p.一次；第0、6和12天)显示出保护颞皮质生长抑素能系统免受Abeta损伤的体内效应。[2]。动物模型：注射Abeta25-35的去卵巢大鼠[2]剂量：300mg给药：腹膜内注射；每天300毫克；结果：恢复的Abeta25-35诱导生长抑素(SRIF)含量和SRIF受体密度降低,并降低SRIF对腺苷酸环化酶活性的抑制作用。
