二十八醇存在于小麦胚芽、甘蔗、苹果、葡萄等的果皮中，主要以脂肪酸酯的形式存在。但在甘蔗中，却存在较多游离态的二十八醇。由上述原料经油脂冬化法、溶剂萃取法、蒸馏法、超临界萃取法等而得。所得粗产品含量不超过50%，粗产品经多次重结晶可得较高纯度产品。
