2-硝基-1，4-二氯苯和4-氨基环己羧酸乙酯反应，再氢化还原，和尿素环合，水解，得到5-氯-1-(哌啶-4-基)-1，3-二氢-2H-苯并咪唑-2-酮。

羟丙胺对2-硝基氯苯中的氯进行亲核取代，再氢化还原硝基为氨基，和氰化钾环合后再氯化，得到1-(3一氯丙基)-2，3-二氢-1H-苯并咪唑-2-酮。

上面得到的两个苯并咪唑衍生物按如下进行，即可得多潘立酮。2.3份1-(3-氯丙基)-2，3-二氢-1H-苯并咪唑-2-酮、2.5份5-氯-l-(哌啶-4-基)-1，3-二氢-2H-苯并咪唑-2-酮、3.2份碳酸钠、0.1份碘化钾和80份4-甲基-2-戊酮一起搅拌，回流24h。冷却至室温，加入水。过滤出不溶物，用硅胶柱进行层析，展开液为氯仿和10%甲醇混合液。收集含产物的流出液，浓缩。剩余物用4-甲基-2-戊酮结晶。过滤后，用二甲基甲酰胺和水的混合液重结晶。得1.3份多潘立酮，收率30%，熔点242.5℃。
