[体外研究]

指南[1](以下是我们推荐的方案。该方案仅提供指南,应根据您的具体需要进行修改)。细胞标记：1.用等渗盐水清洗单层培养细胞,加入或不加入1-5 mM CaCl2或用5 mM Tris-Cl(pH 7.4)缓冲的等渗蔗糖(0.25 M)溶液。2.在室温下,在环境光照条件下,将电池置于适当缓冲液中的Mercyanin 540(1.6 x 10-5 M)中10分钟。3.广泛清洗并用荧光显微镜检查。4.对于原生动物,将0.05 mL含有100-200个生物体的洗涤培养液等分试样放在显微镜载玻片上。5.添加Merocanin 540至最终浓度1.0-10μg/mL。6.用荧光显微镜检查细胞。7.用Merocyanin 540(1.75-5.25 x 10-5 M)培养单细胞悬浮液或组织切片,并按所述用荧光显微镜检查附着细胞。
