丙酮酸脱氢酶可参与到丙酮酸脱羧酶复合物（PDC）中丙酮酸的脱羧和硫辛酸的还原乙酰化。丙酮酸脱氢酶激酶（PDK）可使丙酮酸脱氢酶失活，而丙酮酸脱氢酶磷酸酶则能激活丙酮酸脱氢酶。
