描述 L-同型半胱氨酸(L-HCA)是一种内源性兴奋性氨基酸,作为NMDA受体激动剂(EC50:14μM)。L-同型半胱氨酸具有神经毒性,可用于神经疾病的研究[1][2][3]。

体外研究 L-同型半胱氨酸激活NMDA受体,EC50值为14μM[1]。L-同型半胱氨酸(100μM)在浦肯野细胞中诱导大电流(1.8 nA),该电流对NMDA受体拮抗剂混合物不敏感[1]。L-同型半胱氨酸(250μM,30分钟)在离体鸡胚视网膜中诱导急性兴奋性毒性反应[2]。L-同型半胱氨酸(0-2mM,48小时)在大鼠原代神经元中诱导浓度依赖性神经毒性作用[3]。
体内研究 L-同型半胱氨酸(腹腔注射,4-11mmol/kg)在大鼠出生后早期发育期间诱发癫痫发作[4]。L-同型半胱氨酸(腹腔注射,100-1500mg/kg)部分替代NMDA,在Sprague-Dawley大鼠中,分别在1000和560mg/kg剂量下产生61-67%NMDA水平的最大值[5]。动物模型:Wistar品系雄性白化大鼠[4]剂量:4,5.5,8,11 mM/kg给药:腹腔注射,每日14天结果:以4 mmol/kg诱导屈曲发作。导致剧烈的尾巴摆动、旋转和移动。ECoG(皮层电图)活性下降5-9分钟。
