Luzindole (N-0774) 是一种选择性褪黑素受体 (melatonin receptor) 拮抗剂。Luzindole 优先靶向 MT2 (Mel1b),对于人 MT2 和 MT1 的 Ki 值分别为 10.2 和 158 nM。Luzindole 抑制实验性自身免疫性脑脊髓炎,并发挥抗抑郁样活性。

[体外研究]

Luzindole(N-0774)(5-10μg/ml)抑制MBP反应性LV-4t细胞株的抗原特异性增殖[1]。

[体内研究]

Luzindole(N-0774)(30mg/kg；i.p.；0-5天)抑制实验性自身免疫性脑脊髓炎[2]。Luzindole(N-0774)(30mg/kg i.p.)以剂量依赖性方式减少静止时间,其作用在午夜(减少60%)比中午(减少39%)更明显。Lud吲哚的作用是随时间变化的,在60分钟时表现出最大的效果。通过给予褪黑激素(30 mg/kg Ip),阻止了LuZddol(10 mg/kg Ip)的抗不动作用。Luzindole(30mg/kg i.p.)对白化ND/4小鼠或C57BL/6J小鼠中午或午夜的静止时间没有影响,C57BL/6J小鼠不产生褪黑激素[3]。动物模型：23～12周龄(SJL×PL/J)F1小鼠[2]剂量：30mg/kg给药：i.p.；第0～5天(最小光照条件下23:00～1:00)结果：有效预防实验性自身免疫性脑脊髓炎。
