方法一、醇-醚分离

提取胆固醇和卵磷脂 取新鲜或冷冻大脑或脊髓,去膜及血丝等杂质,绞碎,用1.2倍量丙酮浸渍4.5h,不断搅拌,过滤,反复浸渍5次,滤液制胆固醇。滤饼真空干燥,加入2-3倍量乙醇,于35-40℃提取12h,过滤,再提取1次,滤液制卵磷脂,滤饼真空干燥。

沉淀物的制备 滤饼中加入乙醚提取12h,过滤,再提取1次,合并两次滤液,滤液浓缩,得浓缩液。将浓缩液0℃放置12h,离心,取上清液加入60℃乙醇,边加边搅拌,过滤,收集沉淀物。

滤饼[乙醚]→[12h]滤液[浓缩,离心]→上清液[乙醇]→沉淀物

脑磷脂成品的制备 在沉淀中加入乙醚,0℃静置,过滤,得滤液。滤液中加入乙醇,沉淀,过滤,收集沉淀物,真空干燥,即得脑磷脂成品。

方法二、氯化钙沉淀

粗制脑磷脂的制备 取大脑干加3倍量乙醚,加热至沸,过滤,滤渣再提取一次,合并两次滤液,将滤液浓缩至1/3体积,得浓缩液。

大脑干[乙醚]→滤液[浓缩]→浓缩液

粗制脑磷脂的制备 将浓缩液中加3倍量80%以上的乙醇,水浴加热溶解,冷至过夜,分层,过滤,滤液制卵磷脂和胆固醇,沉淀加少量水,加5g/L。氯化钙,于100℃收集沉淀,将沉淀干燥,得粗制脑磷脂。

脑磷脂成品的制备 粗制脑磷脂加3倍量乙醇,冷室沉淀4h,过滤,如此反复5次,得沉淀,沉淀加乙醚溶解,过滤,得滤液,滤液加等量乙醇或丙酮,沉淀,过滤,将沉淀物真空干燥,即得脑磷脂成品。

粗制脑磷脂[乙醇]→沉淀[乙醚]→滤液[乙醇或丙酮]→脑磷脂成品粗制脑磷脂[乙醇]→沉淀[乙醚]→滤液[乙醇或丙酮]→脑磷脂成品。
