体外研究 Tau聚集和神经炎症-IN-1(化合物30)(0-40μM)降低了30μM下SH-SY5Y细胞的存活率,并且在高浓度下对LO2细胞没有明显的肝毒性,在20μM下对BV2细胞的存活率也没有影响[1]。Tau聚集和神经炎症-IN-1(2.5、5和10μM;24小时)保持松萝酸钠的抗炎活性,并在10μM下抑制LPS刺激的BV2细胞中41%的NO释放速率【1】。细胞毒性试验细胞系:SH-SY5Y、LO2和BV-2细胞【1】浓度:0、10、20、30和40μM孵育时间:24小时结果:30μM降低了SH-SY5Y细胞的存活率,即使在高浓度(高达40μM)下,LO2细胞也没有明显的肝毒性,20μM对BV2细胞的存活率也没有影响。
体内研究 Tau聚集和神经炎症-IN-1(5和10 mg/kg;持续14天)导致OA诱导大鼠的常规参考空间记忆和认知能力显著改善[1]。动物模型:雄性SD大鼠(250-270 g;右背海马微量注射OA)[1]剂量:5和10 mg/kg给药:IP;结果:大鼠常规参考空间记忆和认知能力显著提高。
