葡糖基神经酰胺合酶-IN-2(化合物T-690)是一种有效的脑渗透剂和口服活性葡糖基神经酰胺合酶(GCS)抑制剂,对人GCS和小鼠GCS的IC50分别为15 nM和190 nM。葡萄糖神经酰胺合酶-IN-2与C8神经酰胺和UDP葡萄糖表现出非竞争性抑制。葡萄糖神经酰胺合酶-IN-2可用于高切氏病研究[1]。

体外研究 葡萄糖神经酰胺合酶-IN-2(化合物T-690)没有SERT抑制活性(IC50>10μM)。葡萄糖神经酰胺合酶-IN-2不影响GCase活性(EC50>300μM)[1]。葡糖神经酰胺合酶-IN-2(30μM)不能有效抑制hERG、CaV1。2和导航1。5个频道[1]。
体内研究 葡糖基神经酰胺合酶-IN-2(化合物T-690;po;30100mg/kg)以剂量依赖性方式降低C57BL/6J小鼠血浆和大脑皮层中的GlcCer浓度[1]。葡糖基神经酰胺合酶-IN-2(po;5 mg/kg)的Cmax为416 ng/mL。葡糖基神经酰胺合酶-IN-2表现出良好的口服暴露(BA=31%)[1]。葡萄糖神经酰胺合酶-IN-2显示出良好的大脑暴露(30 mg/kg剂量,1 h时,铜,大脑=0.21μM)[1]。
