| 体外研究 | Rbin-1是真核核糖体生物发生的有效且可逆的基于三唑并吲哚的抑制剂。 Rbin-1抑制重组全长Mdn1的ATP酶活性。两种活性类似物(Rbin-1和Rbin-2)在1μM时抑制ATP酶活性40%。特别地,位置-7处具有溴取代基的类似物(Rbin-2)比Rbin-1活性高10倍(GI50 = 14±1nM(Rbin-2); 136±7nM(Rbin-1) ,n = 4,平均值±SD)[1]。 |
| 激酶实验 | 根据放射性试剂的寿命,将放射性γ-P32-ATP加入体积比为1:1000-1:300的600mM MgATP(pH = 7)溶液中。每个反应的总体积为12mL,包括来自尺寸排阻色谱级分的6ml蛋白质(不同级分的最终浓度为0-50nM,峰级分用于Rbin-1和AMPPNP抑制),4mL FPLC SEC缓冲液。 0.6mM Na 2 SO 4和2mL MgATP(终浓度= 100mM)。然后将反应物在室温下温育30或60分钟,然后用12mL 0.2M EDTA终止反应。将来自每种反应混合物的1mL点在TLC PEI纤维素F板上。 TLC缓冲液含有0.15M甲酸和0.15M氯化锂。然后使用Typhoon Scanner 9400对TLC板成像。使用ImageJ计算对应于放射性游离磷酸盐和ATP的斑点的光密度比,以确定ATP水解的百分比[1]。 |
