| 体外研究 | 八聚体结合转录因子4(Oct4)是诱导和维持细胞多能性的主要调节因子,并且在分化的早期阶段具有关键作用。它是唯一不能被相同蛋白家族的其他成员取代以诱导多能性的因素[1]。 Oct4已被证明是胚胎干细胞(ESC)多能性的重要调节因子,也是重编程过程的关键。 OAC2是OAC1的结构类似物。 OAC2以与OAC1相似的程度激活Oct4和Nanog报告基因。 OAC1及其两种结构类似物OAC2和OAC3将重编程效率提高四倍,高达2.75%,并且当与四种重编程因子Oct4,Sox2,Klf4和四种重编程因子组合使用时,加速iPSC集落的出现3至4天。 c-Myc的[2]。 |
| 细胞实验 | 用化合物OAC1或其结构类似物OAC2,OAC3以1μM浓度或指定浓度处理Oct4-luc或Nanog-luc细胞。使用的其他化合物包括2μMBIO,2μMBIX,2μM5'-氮杂胞苷,25μg/ mL维生素C,10nMAm580,5μM反苯环丙胺和0.5mM valporic acid。荧光素酶报告基因测定在化合物处理后24小时或在指定的时间点进行。对于Topflash报道分析,使用转染将0.2μgβ-连环蛋白反应性Topflash报告基因质粒引入CV1细胞。转染后6小时加入化合物。使用Glo Luciferase Assay System [2]在化合物处理后48小时测量荧光素酶活性。 |
