In Vivo ABBV-4083 (150 mg/kg, once daily for 14 days) reduces Wolbachia levels (measured 16 weeks post-treatment-initiation, pti) in mice by >99.9% in the recovered female adult worms. Levels of circulating microfilariae declines from ~7 weeks pti and are completely cleared from 12 weeks pti until the end of this study at 16 weeks pti[1]. Animal Model: Female BALB/c mice or female jirds were infected at 6–8 weeks of age with L. sigmodontis larvae through the bites of Ornithonyssus bacoti mites[1]. Dosage: 150 mg/kg. Administration: PO once daily for 14 days started at 14 weeks post infection (pti). Result: Wolbachia levels (measured 16 weeks post-treatment-initiation, pti) were reducedby >99.9% in the recovered female adult worms. Starting at ~7 weeks pti, levels of circulating microfilariae declined and were completely cleared from 12 weeks pti until the end of this study at 16 weeks pti.
