环糊精是1891年由Villers发现的，本世纪初又分离成功α及β环糊精。1972-1973年制造环糊精酵素（即环糊精葡糖转位酶，简称CGT-ase）的细菌发现，特别是其中的嗜碱性细菌，对于大规模生产环糊精具有十分重大的意义。以马铃薯、玉米、小麦等加工的淀粉为原料，经下列过程可得环糊精：淀粉悬浮液[70 -80 ℃ 液化][ 60 ℃ 与乙醇反应]、加热失活、过滤、浓缩干燥。
