[体外研究]

间隙27导致在牛骨切片上培养的TRAP阳性单核和多核大鼠破骨细胞的数量显着减少。通过测量所有TRAP阳性细胞的肌动蛋白环的破骨细胞百分比,发现剩余破骨细胞的活性减少。此外,处理过的培养物中的再吸收区域大大减少[1]。用间隙连接抑制剂Gap 27(500μM)孵育颈动脉基本上消除了对乙酰胆碱的超极化,但对左旋克罗卡林没有影响。在豚鼠分离的颈内动脉中,Gap 27抑制乙酰胆碱诱导的内皮依赖性超极化[2]。

[体内研究]

间隙27(300μM)抑制胸主动脉和肠系膜上动脉的松弛40％。间隙27还减弱胸主动脉中ATP诱导的松弛的内皮依赖性组分。 [3]。

[细胞实验]

用三种不同的处理（对照，庚醇和Gap 27）培养骨细胞培养物48小时。培养期后，固定骨片。对细胞进行抗酒石酸酸性磷酸酶（TRAP）染色。为了使细胞核可视化，将细胞与结合DNA的荧光染料Hoechst 33258（在PBS中1：800稀释的1mg / mL储备液）在室温下孵育10分钟。计算每个骨切片上单核和多核TRAP阳性细胞的数量[1]。
