由1,4-丁二醇与环氧氯丙烷缩聚制得。

乙二醇二缩水甘油醚的制法有多种，如一步法、两步法、烯烃氧化法、相转移催化法等。两步法　以二元醇与ECH为主原料，在路易斯酸催化下［一般采用BF3·O（CH3CH3）2催化剂］开环加成，然后在碱的作用下闭环，得到二缩水甘油醚。反应式为：

制备实例：在装有搅拌器、回流冷凝管的500mL三口瓶中加入定量的乙二醇和催化剂，开动搅拌器，加热到一定温度后，开始由恒压漏斗滴加ECH，加完后恒温一段时间，加入一定量的溶剂和碱，再反应一段时间后，抽滤，减压脱除溶剂得产物。

相转移催化法　称取148g环氧氯丙烷、60～70g氢氧化钾，加少许水和一定量的相转移催化剂（二苯并-18-冠醚-6、三甲基苄基氯化铵、正四丁基溴化铵等），快速搅拌，在45℃以下滴入10g乙二醇，再在40℃下继续反应60min。过滤，残渣用二氯甲烷洗两次；滤液干燥后，首先常压蒸馏，除去过剩的环氧氯丙烷和溶剂；再改为减压蒸馏，在799.93Pa下收集118～121℃的馏分。得产品19.7g。
