异丙隆制备

1、 由对异丙基苯异氰酸酯与二甲胺反应而得。将33%的二甲胺水溶液加入反应瓶,加水,冷却至0-5℃时,加速搅拌,滴加对异丙基苯异氰酸酯。在0-5℃下搅拌反应2h,抽滤,将滤饼在丙酮中重结晶即得成品。原料消耗定额:对异丙基苯胺(95%)980kg/t、二甲胺(40%)950kg/t、光气(80%)840kg/t、氯气570kg/t。

2、 异氰酸酯法
异氰酸异丙苯酯的制备 以甲苯为溶剂,4-异丙基苯胺与光气作用生成异氰酸异丙苯酯。
异丙隆的合成 异氰酸异丙苯酯与二甲胺加及生成异丙隆。投料经为异氰酸酯:二甲胺=1:1 (mol比);滴加温度15℃,时间20min;反应温度20~30℃,时间2h。以水为溶剂,收率95%。
氨基甲酰氯法
先制备N,N-二甲基氨基甲酰氯的制备; 再与4-异丙基苯胺作用合成异丙隆。
非光气法
对异遭受基三氯乙酰苯胺的制备 将13.5g (0.1mol)对异丙基苯胺和16.5g (0.1mol)三氯乙酸混合后,搅拌下滴加14.0g (0.1mol)三氯化磷,温度控制在100~110℃,滴毕恒温反应30min,然后加入100mL冰水,抽滤收集固体并用水洗至中性,用乙醇/水重结晶,得白色结晶,收率91%, m.p.123~125℃。
异丙隆的合成 将14.0g (0.05mol)对异丙基三氯乙酰苯胺、5.0g(0.125mol)氢氧化钠、4.5g (0.10mol)二甲胺和80mL二甲苯亚砜加入反应器中,于60~80℃反应30min,反应毕倾入水中,抽滤收集固体,用乙醇/水重结晶,得白色固体,收率95%,m.p.152~154℃。
据Atanassova, I.A.报道,此反应过程可能是对异丙基三氯乙酰苯胺在碱的作用下,首先生成中间体对异丙基异氰酸苯酯,然后立即与二甲胺作用生成异丙醇。
采用非光气法合成异丙隆,还可用尿素代替光气,在水溶液中与对异丙基苯胺反应,生成中间体对异丙基苯脲,然后再与二甲胺水溶液反应,合成异丙隆,总收率达76%。
