3'-Sialyllactose(3'-SL)是一种益生元,维持免疫稳态,发挥抗炎和抗关节炎作用。3'-唾液乳糖可用于炎症研究[1][2]。

[体外研究]

3'-Sialyllactose(0-250μM；24-36 h)促进和恢复Col2a1合成,积累细胞外硫酸化蛋白聚糖,并抑制炎症细胞因子的作用[1]。3'-Sialyllactose(0-250μM；24小时)激活软骨细胞中Sox9的表达并抑制NF-κB的激活[1]。Western Blot分析[1]细胞系：软骨细胞浓度：0、50、100和250μM孵育时间：24-36小时结果：IL-1β处理的软骨细胞中,Col2a1转录和蛋白水平随剂量增加而增加,Col2a表达恢复。剂量依赖性抑制IL-1β诱导的软骨细胞中Mmp3、Mmp13和Cox2的表达。IL-6、IL-17和TNF-α诱导软骨细胞中Mmp3、Mmp13和Cox2表达降低。

[体内研究]

3'-Sialyllactose(10-100 mg/kg；口服,每周三次,共6周)可保护小鼠免受骨关节炎软骨破坏[1]。动物模型：采用内侧半月板手术的8周龄雄性C57BL/6小鼠[1]剂量：10、50和100 mg/kg给药：口服灌胃；10-100 mg/kg,每周三次；结果：通过分解代谢因子的表达,有效保护骨关节炎小鼠免受软骨破坏。
