[体外研究]

在体外实验中,溴化氟托溴铵比阿托品更有效[3]。

[体内研究]

溴化氟托溴铵(0.0003%;单次吸入)抑制乙酰胆碱诱导的支气管收缩,而不会改变豚鼠的血压下降[1]。溴化氟托溴铵(3和10 mg/kg;静脉注射)显示出弱的肥大细胞稳定作用,但对LTD4和5-羟色胺没有拮抗作用[2]。动物模型:豚鼠[2]剂量:3和10 mg/kg给药:静脉注射;结果:显示对48小时同源PCA具有抑制作用,并抑制豚鼠抗原刺激的分离的大鼠肥大细胞释放组胺。

氟托溴铵制备

以托品为原料。
38.9g(0.24mo1)N,N’-羰基二咪唑溶于二氯甲烷,在15min内,加入54.8g(0.24mo1)无水羟基二苯乙酸,于15~20℃混合。当羟基二苯乙酸溶解,而羟基二苯乙酸咪唑化物缓慢以结晶的形式沉淀出。反应1h,过滤出该沉淀,用50ml二氯甲烷洗,真空70℃干燥,得64.7g粗品固体,收率96.8%粗品熔点138~139℃(分解)。经丙酮重结晶后,熔点147~148℃(分解)。将27.8g(0.1mol)上述固体分成四份,每次间隔15min,加到14.1g(0.1mo1)无水托品在100ml无水丙酮的沸腾溶液中。当加到第四份时,羟基二苯乙酸托品酯以粗晶体的形式沉淀出来。在沸腾的溶液中继续反应2h,然后冷至0℃。过滤该沉淀,用0℃的丙酮洗,真空70℃干燥,得30.5g羟基二苯乙酸托品酯,收率86.8%,熔点146~147℃。
羟基二苯乙酸托品酯经高锰酸钾氧化去甲基化,得到羟基二苯乙酸去甲托品酯。
146.0g(0.434mo1)羟基二苯乙酸去甲托品酯、60.6g(0.477mo1)2-溴氟乙烷和101.1g(0.954mo1)碳酸钠在1200ml乙腈中,回流搅拌10h。蒸去乙腈,剩余物溶于水和二氯甲烷。分出的碱性水溶液用二氯甲烷重复提取。二氯甲烷层合并,无水硫酸钠干燥。过滤,按常法通氯化氢可得166.3g羟基二苯乙酸N-β-氟乙基去甲托品酯盐酸盐,收率91.5%,甲醇-乙醚重结晶得白色结晶,熔点209℃(分解)。
109.9g(0.287mo1)羟基二苯乙酸N-β-氟乙基去甲托品酯(按常法从其盐酸盐得到)溶于450ml无水二氯甲烷和300ml无水乙腈的混合液中,加入136.1g(1.433mo1)溴甲烷,在室温季铵化。3天后,过滤收集结晶,用丙酮洗,于1.6kPa真空和50℃干燥,得129.8g白色结晶的氟托溴铵,收率94.7%。以乙腈重结晶:熔点192~193℃(分解);以乙醇-丙酮重结晶:熔点198-199℃(分解)。
