由4-氯-2-硝基苯胺在苯或甲苯溶剂中、在催化剂吡啶存在下与双乙烯酮反应,生成相应的α乙酰化物,该产物经碱性水溶液处理,生成相应的喹喔啉酮氮氧化物。喹喔啉酮氮氧化物在氢氧化钠(或氢氧化钾)存在下,以甲苯水为溶剂,于60~70℃反应,生成的氮氧化物在碱水溶液中,于室温下,与一定量的硼氢化钠反应,得6-氯-2(1H)喹喔啉酮。6-氯-2(1H)-喹喔啉酮在DMF溶液中与氯化亚砜作用,得2,6-二氯喹喔啉。然后在氮气保护下,在DMF溶剂中,2,6-二氯喹喔啉与对苯二酚缩合可得到4-(6-氯-2-喹喔啉氧基)酚。4-(-6-氯-2-喹喔啉氧基)酚与α-卤代丙酸乙酯在乙腈溶液中,以碳酸钾为缚酸剂,缩合得到喹禾灵。喹禾灵也可用2,6-二氯喹喔啉与α-对羟基苯氧基丙酸乙酯在碳酸钾存在下,于乙腈溶液中回流直接作用制得。
