[体外研究]

添加聚山梨酯20可以改变木质素的表面性质,改变其疏水性、氢键能力和表面电荷,从而减少蛋白质的非生产性吸附[1]。将聚山梨酯20(0.35%-0.7%w/w)添加到0.05 mol/L pH 6.5的磷酸盐缓冲液中,可显著增加药物的表观溶解度,从0.8μg/ml甚至增加到353μg/ml,而在较低pH值或水中的效果则小得多(pH 4.5中为20μg/ml)。在pH值为6.5、含有0.7%聚山梨酯20的情况下,磷酸盐浓度(0.2 mol/l)增加,导致坎地沙坦-西来塞替尔的溶解度进一步增加至620μg/ml[2]。

吐温20制备

1.将1 mol斯盘20投入反应釜中,在搅拌下加50%液碱作催化剂,逐渐升温,减压脱水,把水脱尽后用氮气置换釜中的空气,驱净空气后升温至120℃开始通入20 mol环氧乙烷,反应温度维持在150~170℃,压力0.2~0.3 MPa。通完环氧乙烷后,冷却至80~90℃,将料液打入中和釜用冰醋酸中和,再用双氧水脱色,最后冷却脱色,最后冷却放料,包装即为成品。在甲醇钠(或乙醇钠)的催化下,在130~170℃时,在span-20中通入一定量的环氧乙烷发生加成反应得产品。
