| 体外研究 | 14-脱氧-11,12-二脱水穿心莲内酯,一种天然存在的穿心莲内酯的非细胞毒性类似物,有效地减少卵清蛋白(OVA)诱导的炎性细胞募集到支气管肺泡灌洗液(BAL),IL-4,IL-5,IL-13和嗜酸性粒细胞趋化因子的产生,血清IgE合成,肺嗜酸性粒细胞增多,粘液分泌过多,肥大细胞脱颗粒和小鼠哮喘模型的气道高反应性(AHR),可能是通过抑制NF-κB活性[1]。 |
| 体内研究 | 14-脱氧-11,12-二脱水穿心莲内酯(1mg/kg)显着降低OVA攻击的小鼠对乙酰甲胆碱的抗性(R1)和恢复Cdyn [1]。 |
| 细胞实验 | 将A549细胞(3×103 /孔),BEAS-2B细胞(5×103 /孔)和RBL-2H3细胞(3×103 /孔)接种在平底96孔板中过夜,然后孵育增加浓度(3-120μM)的14-脱氧-11,12-二脱水穿心莲内酯或穿心莲内酯在37℃下24和48小时。使用CellTiter 96 AQueous细胞增殖测定分析细胞活力。该MTS测定基于活细胞将可溶性四唑盐转化为有色甲product产物的能力。吸光度记录在490 nm [1]。 |
| 动物实验 | 小鼠[1]将6至8周龄的雌性BALB / c小鼠致敏并用OVA攻击。简而言之,在第0天和第14天,通过腹膜内注射20μgOVA和4mg悬浮在0.1mL盐水中的Al(OH)3致敏小鼠。在第22,23和24天,用1%OVA攻击小鼠。气溶胶30分钟。在每次OVA气溶胶攻击之前2小时和之后10小时通过ip注射给予在0.1mL盐水中的14-脱氧-11,12-二脱水穿心莲内酯(0.1,0.5和1mg / kg)或载体(1%DMSO)。盐水气溶胶用作阴性对照。 |
