制备

将对甲氧基苯乙腈溶于四氢呋喃中，在氮气保护加入丁烷锂的己烷溶液。再加入环己酮进行反应。反应液放入含有冰的氯化铵饱和水溶液，再加入水。过滤沉淀，用水和乙醚洗，得1-[氰基(4-甲氧苯基)基]环己醇。将其溶于氨乙醇混合液，加入铑氧化铝进行催化氢化。滤去催化剂，用乙醇洗。洗液和滤液合并，浓缩至油状物。将该油状物、甲醛水溶液、甲酸和水一起搅拌过夜。反应液浓缩，加入水，用浓盐酸调至pH为2，用乙酸乙酯提取以移去粉红色的杂质。提取后的水溶液用氢氧化钠溶液碱化，然后用乙酸乙酯提取。提取液用饱和盐水洗，干燥、过滤、浓缩。得到的物质溶于乙酸乙酯，加入氯化氢的异丙醇溶液，收集得盐酸文拉法辛。将消旋的文拉法辛溶于乙酸乙酯中，加入二(对甲苯基)-L-酒石酸单水合物的乙酸乙酯溶液，进行拆分，得到光学异构体的(+)或(-)文拉法辛，酸化得到盐酸盐。
