NF-κB

体外研究 Flaconitine已被证明可抑制TNF-α诱导的NF-κB活化,被认为是二酯-二萜型NF-κB抑制剂[1] Flaconitine(3-Acetylaconitine)用于评估其对人白血病P388细胞的细胞毒活性。 SRB(Sulforhodamine B)方法,以依托泊苷为阳性对照。 Flaconitine的细胞生长抑制效力表示为IC 50(μM)值。 Flaconitine具有中等细胞毒活性,IC50为18.6μM[2]。
细胞实验 按照标准公开的程序,使用微量滴定板形式和磺胺罗丹明B在终点进行测定。将白血病细胞(P388)暴露于乌头碱(3-乙酰基乌头碱)三天。使细胞生长并保持在设定为37℃,5%CO 2和95%湿度的培养箱中。补充有10%胎牛血清(FCS)和0.5%胰蛋白酶的RPMI-1640培养基用于培养P388细胞[2]。
