描述 STAT3-IN-14(化合物1)是STAT3抑制剂,具有STAT3磷酸化抑制活性。STAT3-IN-14(化合物1)可以直接结合到STAT3的铰链区[1]。

体外研究 STAT3-IN-14(化合物1)显示出对磷光体-STAT3的抑制活性。STAT3-IN-14以DOE依赖性方式降低了对MDA-MB-231细胞的磷酸化抑制活性(JAK/STAT途径组成性活性),但对JAK激酶没有抑制活性。STAT3-IN-14(化合物1)具有生长抑制活性,MDA-MB-231、MCF7和A549细胞的IC50值分别为0.42μM、0.50μM和0.13μM[1]。Western印迹分析[1]细胞系:MCF7细胞;MDA-MB-231细胞浓度:1或10μM;0.5、1、2、4μM孵育时间:1h;2h结果:10μM时完全抑制STAT3磷酸化,1μM时部分抑制。以DOE依赖性方式降低STAT3的磷酸化。细胞增殖试验[1]细胞系:MDA-MB-231、MCF7和A549细胞浓度:0-30μM孵育时间:72 h结果:三种癌细胞系中均表现出中等至强的生长抑制活性。
