体外研究	CFSE是一种荧光染料,可以跟踪细胞分裂[1]。为了检查免疫细胞迁移到活切片中的能力,分离自体脾细胞或PBMC的单细胞悬浮液,用CFSE标记它们,并在培养的第2天将它们添加到切片的顶部。再培养6天后,发现CFSE标记的免疫细胞(绿色)已经在整个切片中迁移。没有添加CFSE标记的免疫细胞,切片中未发现阳性细胞[2]。
细胞实验	将脾细胞（或PBMC）的单细胞悬液用PBS中的1μMCFSE在37℃下染色9分钟，在室温下与20mL 10％FBS RPMI-1640培养基一起染色2分钟，离心，洗涤并计数。将2×106个CFSE标记的脾细胞加入到24孔膜培养插入物中的切片顶部[2]。
