| 体外研究 | D5261(0-10µM,72 h)在Ba/F3 MPRIP TrkA细胞(IC50=3.32µM)和Ba/F3 CD74 TrkA细胞(IC50=2.91µM)中显示出良好的抗增殖活性[1]。D5261在一定程度上表现出对TrkB/C的细胞选择性(Ba/F3 IC50>10µM,Ba/F3 QKI TrkB IC50=6.17µM,Ba/F3-EML4-TrkC IC50>10µM)[1]。D5261(0-50µM)以浓度依赖性方式抑制TrkA自身磷酸化和下游信号传导[1]。D5261对含有TrkA突变体(G667C、G667S、G667A、G595R和V573 M)的Ba/F3细胞具有抑制活性【1】。细胞活性测定【1】细胞系:Ba/F3 MPRIP TrkA,Ba/F3 CD74 TrkA细胞浓度:0-10µM培养时间:72 h结果:对Ba/F3 MPRIP TrkA和Ba/F3 CD74 TrkA细胞显示出良好的抗增殖活性,IC50分别为3.32和2.91µM。Western Blot分析【1】细胞系:Ba/F3 CD74 TrkA细胞浓度:0.08、0.4、2、10和50µM培养时间:结果:以浓度依赖性方式抑制TrkA自身磷酸化和下游PLCγ1、AKT和ERK的磷酸化。 |
