• 体外研究: 暴露于1μMTPOP146导致恢复半衰期显着降低,这与含有溴结构域失活突变N1168F的构建体相当,证明TPOP146靶向细胞核中的CBP溴结构域并且能够与乙酰基-赖氨酸介导的相互作用竞争。 CBP溴结构域在细胞环境中的作用[1]。
• 激酶实验: 将蛋白质缓冲在10mM HEPES，pH 7.5,500mM NaCl中，并在96孔板中以20μL体积中的终浓度2μM进行测定。加入化合物（TPOP146），终浓度为10μM。以1：1000的稀释度添加SYPRO Orange（Molecular Probes）作为荧光探针。 SYPRO Orange染料的激发和发射滤光片分别设定为465和590 nm。在25至96°C的温度范围内以每分钟3°C的速度升温，并在每个间隔[1]进行荧光读数。
