TIM-063(TIM063)是一种有效、选择性、ATP竞争性的CaMKK抑制剂,对CaMKKα和CaMKKβ的Ki分别为0.35/0.2μm。TIM-063直接针对CaMKK的催化结构域,类似于STO-609。TIM-063在IC50为0.3μm的HeLa细胞中抑制离子霉素诱导的外源表达的CaMKI、CaMKIV和内源性AMPKα的磷酸化。TIM-063抑制转染COS-7细胞中CaMKK亚型介导的CaMKIV磷酸化。TIM-063显示细胞通透性和抑制细胞内CaMKK活性的能力。TIM-063是一种有用的化学探针,用于精确分析CaMKK介导的信号通路。
