麦芽酚制备

1.麦芽醇有三种主要制法:1.从天然产物中提取 麦芽醇存在于小麦、大豆的分解液中和大麦焙烧凝缩物中,也存在于某些枞树和落叶松树皮层中。1986年美国就用萃取结晶法从木醋中提取麦芽醇。1970年美国又从落叶松树皮中用氯仿提取麦芽醇。日本从连香树叶中用氯仿直接浸提或从其水蒸气蒸馏液中提取。从木材干馏所得的木焦油进行减压蒸馏也可分离到麦芽醇。2.半合成法(以曲酸为原料)曲酸的母体是葡萄糖,相当于3-酮-葡萄糖的脱水产物。将淀粉用黄曲霉菌发酵,在发酵液中产生的曲酸,经转变成曲酸把催化下用空气(或氧气)氧化成靠闷酸,然后在225℃下脱羧成焦袂康酸。在碱性条件下将焦袂康酸与甲醛反应,生成羟基麦芽醇。然后用盐酸和锌粉进行还原得麦芽醇。经蒸馏或升华,在氯仿中重结晶,即得麦芽醇成品。从曲酸制成靠闷酸,也可通过曲酸与氯化苄经醚化生成曲酸苄醚,再用二氧化锰氧化成靠闷酸醚,然后脱苄基生成靠闷酸。3.用糖醛为原料合成麦芽醇。

2.糠醇法
糠醇在甲醇水溶液中通人氯气氯化生成4-氯代-6-羟基-2[H]-吡喃-3[6H]-酮,然后加热水解得焦袂康酸,在碱性下将其与甲醛缩合得羟甲基焦袂康酸,用锌粉在盐酸中将其还原为麦芽酚。半合成法
以淀粉为原料,经梨头霉发酵制得曲酸。曲酸与氯化苄在氢氧化钠-甲醇溶液中醚化生成曲酸苄醚;然后用二氧化锰氧化生成考门酸苄醚,并在盐酸存在下脱苄生成考门酸,再脱羧成为焦袂康酸,最后与甲醛化合,并用锌粉还原成麦芽酚。
糠醛法
糠醛与甲基溴化镁作用得到甲基糠醇,然后在甲醇水溶液中、0℃下通氯气氧化,接着加热至100℃水解得到麦芽酚。此法原料来源广,工艺路线短,条件温和,设备简单,“三废”少,有较好的发展前景。

由发酵法制得曲酸后与氯化苄进行醚化得曲酸苄醚,经二氧化锰和盐酸催化下生成考闷酸,再经脱羧得焦炔康酸,经甲基化而成。
用氮杂环己烷反应成焦炔康酸,然后在2位甲基化后制成。
乙二酸二乙酯在乙醇钠存在下与丙酮缩合后在三氯甲烷溶液中通入溴,得γ-衍生物;再由氢氧化钾进行加水分解成袂康酸,然后加热脱羧基得焦袂康酸,用甲醛和哌啶处理再还原而得。

3.将曲酸在钯催化下用空气 ( 或氧气)氧化成靠闷酸,再高温脱羧成焦羨康酸。在碱性条件下与甲醛反应,生成羟基麦芽醇。然后用盐酸和锌粉进行还原得麦芽醇。经蒸馏或升华,在氯仿中重结晶,即得麦芽醇精品。
