AACOCF3(花生四烯酸三氟甲基酮)用途

AACOCF3(花生四烯基三氟甲基酮)是花生四烯酸的细胞渗透性三氟甲基酮类似物。AACOCF3是85 kDa细胞溶质磷脂酶A2(cPLA2)的有效和选择性慢结合抑制剂。AACOCF3阻断钙离子载体激发血小板产生花生四烯酸和12-羟基二十碳四烯酸。AACOCF3抑制葡萄糖诱导的大鼠胰岛胰岛素分泌。AACOCF3具有心血管疾病研究的潜力[1][2][3]。

[体外研究]

AACOCF3抑制钙离子载体激发的U937细胞(IC50=8μM,2 x 106细胞ml-1)和血小板(IC50=2μM,4 x 107细胞ml-1)释放花生四烯酸[1]。AACOCF3(10μM)抑制HAOSMC中磷酸盐诱导的钙化和成骨/软骨信号传导。AACOCF3显著抑制基础和Pi诱导的花生四烯酸释放,花生四烯酸是PLA2活性的产物[2]。

[体内研究]

AACOCF3(10 mg/kg；灌胃；每周5天；ApoE–/–喂食高胆固醇饮食的小鼠(6周龄雄性)显著降低III型胶原斑块的表达,但对总胶原积累没有显著影响[3]。
