由丙二酸二乙酯与硫脲反应而得。将硫脲溶于甲醇中，加入甲醇钠及丙二酸二乙酯，加热回流4-5h，然后减压蒸馏回收甲醇至尽，冷却后加入盐酸酸化至pH为1-2。放置过夜，析出硫代巴比妥酸结晶，过滤，用水重结晶即为纯品。
