胸腺五肽制备

胸腺生成素-II的制备。
原料处理、浓缩以小牛胸腺为原料，加入0.1mol/L NH4HCO3，搅匀提取，得碱性蛋白液，加热至70℃除去变性杂蛋白，去除沉淀，将上清液浓缩。
小牛胸腺[0.1 mol/L NH4HCO3]→碱性蛋白液[70℃, 去杂蛋白]→[浓缩]浓缩液
分离、层析、精制经SephadexG-50和羟基磷石灰柱层析，分离，再经QAE-Sephadex柱层析，精制，即得胸腺生成素-II
浓缩液[Sephadex G-50]→[羟基磷石灰柱]层析液[QAE-Sephadex 柱]→胸腺生成素-II
胸腺生成素-II的提取
原料处理、超过滤、离心以新鲜牛脾为原料，加入0.1 mol/L NH4HCO3，匀浆提取，得碱性可溶性蛋白。用H10×100超过滤，将滤液浓缩，水浴加热至70℃，离心15 min，留上清液。
新鲜牛脾[0.1 mol/L NH4HCO3]→碱性蛋白液[超过滤，浓缩]→[70℃，离心15min]上清液
过柱纯化取上清液调节pH为5.2，加入阳离子交换树脂(50 mg/ml)，搅拌，过滤，再将滤液调节pH为9.5，加入阳离子交换树脂(50mg/m1)，搅拌，过滤，得滤液。
上清液[pH5.2, 阳离子柱]→滤液[pH9.5, 阳离子柱]→滤液
凝胶滤过、纯化滤液经Sephadex G-75凝胶过滤，得活性TP和泛激素，再经阴离子交换树脂层析(NH4HCO3, 离子强度变化0.1-0.8mol/L)，得 TP-III、杂蛋白和泛激素。
滤液[Sephadex G-75凝胶过滤]→活性TP和泛激素[阴离子树脂层析]→得TP-III、杂蛋白和泛激素。
