[体内研究]

达特罗班(BM-13505)(1毫克/千克；静脉注射；每小时)对猫急性心肌缺血后再灌注损伤有保护作用[2]。与溶媒(生理盐水)处理的猫相比,达曲班(20 mg/kg/h静脉注射)减少了缺血引起的ST段抬高,并阻止了再灌注期间心电图Q波的发展。达特罗班保护心肌免受缺血损伤,这种作用涉及预防缺血诱导的白细胞增多[3]。动物模型：成年雄性猫(2.8～4.6kg；麻醉猫模型)[2]剂量：1 mg/kg给药：静脉注射；再灌注前30分钟,1 mg/kg,1 mg/kg/小时。结果：在不改变基本血流动力学的情况下,缺血组织面积占总左心室重量和总危险面积的百分比显著减少从而不影响心肌耗氧量。

4-氰甲基苯乙酸溶于无水乙醇，加入氯化钯和浓盐酸，在室温和0.5MPa通氢。滤除催化剂，滤液减压蒸出溶剂。加入水，充分溶解，过滤，滤液为化合物(I)的溶液。往该溶液中，加入碳酸钾，再在80℃加入对氯苯磺酰氯。加毕，在80℃搅拌。冷却，用2mol/L盐酸酸化至Ph值2。滤集固体，水洗，干燥。再用66%乙醇重结晶，得达曲班，熔点138～140℃。
