胰激肽释放酶制备

1.猪胰脏脱水↓丙酮提取↓乙酸沉淀↓丙酮溶解↓NaCl,氨水沉淀↓丙酮

离子交换↓乙酸缓冲液洗涤↓乙酸缓冲液洗涤↓NaCl溶液透析，冻干精品

将新鲜猪胰脏绞碎，用丙酮浸泡脱水，制成猪胰丙酮粉，然后加20倍量002mol/L醋酸，于10℃搅拌提取12h，离心，残渣加10倍量002mol/L醋酸再提取6h。提取液合并，然后加冷丙酮至浓度达33%，过滤，滤液再补加冷丙酮至70%，静置4h，离心，收集沉淀。沉淀再用丙酮、乙醚脱水、脱脂，真空干燥，得血管舒缓素粗品。向血管舒缓素粗品中加50倍量02%冷氯化钠溶液，用氨水调pH=80，搅拌溶解后过滤，滤液应澄清。将澄清滤液冷至2～3℃，再加冷丙酮使浓度达40%，冷室静置过夜，离心。离心清液补加丙酮至浓度为60%，静置4h，离心分出沉淀。沉淀用丙酮、乙醚洗涤，真空干燥，得血管舒素中间品。将中间品溶于0001mol/L、pH=45醋酸缓冲溶液中，离心去除沉淀。清液中加入钠型弱酸性阳离子树脂AmberliteCG50（树脂∶中间品=50∶1），搅拌吸附2h，吸附后的树脂用0.001mol/L、pH=45缓冲液洗涤后，用2倍1mol/L、pH=50氯化钠溶液搅拌洗脱1h，分离树脂。洗脱液透析脱盐，冻干，即得血管舒缓素。
