[体外研究]

当细胞色素c和dATP在NS3694(10-100μM；120 min)存在下刺激细胞提取物时,可观察到对caspase 9、3和7处理、DFF45/ICAD和PARP裂解以及脱辅酶活性的剂量依赖性抑制[1]。NS3694(10-100μM)抑制TNF诱导的MCF-casp3细胞效应物caspase激活和凋亡[1]。NS3694特异性抑制细胞色素C诱导的凋亡体复合物形成和caspase-9激活,从而抑制线粒体介导的凋亡。NS3694通过阻断启动子caspase-9的激活来抑制凋亡体Apaf-1的形成[1]。Western Blot分析[1]细胞系：HeLa细胞浓度：10μM、50μM、100μM孵育时间：120分钟结果：抑制细胞色素c-和dATP诱导的脱羧酶激活和半胱天冬酶和半胱天冬酶底物的处理。
