• 体外研究: PF-05231023是一种长效成纤维细胞生长因子21(FGF21)类似物[2]。
• 体内研究: 给予天然FGF21和PF-05231023(3mg/kg或10mg/kg)可改善Zucker大鼠的葡萄糖耐量和肝脏胰岛素敏感性[3]。 PF-05231023(10 mg/kg)对秋田视网膜有保护作用。 PF-05231023(10mg/kg)降低糖尿病小鼠的视网膜炎症。施用PF-05231023(10mg/kg)减少Akita小鼠中的视网膜IL-1βmRNA表达。施用PF-05231023(10mg/kg)增加秋田小鼠中锥体特异性抑制蛋白4的表达。 PF-05231023(10mg/kg)给药恢复Akita小鼠的光感受器形态。 PF-05231023(10mg/kg)抑制光感受器中的氧化应激诱导的炎症。施用PF-05231023(10mg/kg)可降低视网膜NRF2水平的变异性,并显示秋田小鼠中NRF2水平升高的趋势(非显着)[4]。
