• 体外研究: Wnt-C59(C59)在纳摩尔浓度下体外抑制PORCN活性,如通过抑制Wnt棕榈酰化,Wnt与载体蛋白Wntless/WLS的相互作用,Wnt分泌和β-连环蛋白报道活性的Wnt活化所评估的。 Wnt-C59抑制WNT3A介导的多聚化TCF结合位点的激活,驱动荧光素酶,IC50为74 pM [1]。
• 体内研究: Wnt-C59在小鼠中显示出良好的生物利用度。 Wnt-C59阻断MMTV-WNT1转基因小鼠乳腺肿瘤的进展,同时下调Wnt /β-连环蛋白靶基因[1]。 Wnt-C59具有消除人类肿瘤中癌症干细胞的潜力。 Wnt-C59通过阻止HNE1和SUNE1细胞中的球形成,以剂量依赖的方式抑制NPC细胞的干性[2]。
• 细胞实验: 将1×104 HK1，CNE1，HNE1和SUNE1细胞接种在24孔板中，第二天加入Wnt-C59（5μM，10μM和20μM）。在接种细胞后24,48,72和96小时通过显微镜确定细胞汇合。 IC50由MTT测定法确定[2]。
• 动物实验: 小鼠[1]用载体或Wnt-C59 10mg / kg每天一次处理用独立MMTV-WNT1肿瘤原位移植的雌性裸鼠，持续17天。肿瘤体积在隔天测量[1]。
