体外研究	将培养的MIN6细胞与无血清培养基(SFM)(对照),1μMExareorelin(Hex),1mM链脲佐菌素(STZ)单独或STZ孵育,然后用Examorelin孵育6小时。 MIN6的活力受细胞不同处理的显着影响(F(3,36)= 5.244,P <0.01)。与仅用STZ治疗的患者相比,用STZ处理的MIN6细胞接着是Examorelin,细胞活力大大增加(对照组的105.3％,P <0.01)[2]。
体内研究	Examorelin(Hexarelin)治疗可改善心脏收缩功能,减少丙二醛产生,并增加存活的心肌细胞数量。 Examorelin治疗的有益效果略微优于等摩尔ghrelin治疗。 Examorelin还诱导I/R心肌中IL-1β表达的下调和IL-1Ra表达的上调,其可被GHSR拮抗剂(D-Lys3)-GHRP-6中和。 Examorelin部分通过激活心脏GHSR1a受体修饰IL-1信号通路,保护体内心肌细胞免受I/R损伤[1]。检查了Examorelin(100μg/ kg)在STZ诱导的糖尿病大鼠中的作用。 STZ注射显着增加大鼠的血糖水平,而Examorelin(Hex)治疗减少了STZ的效果。分别在禁食和进食条件下测量大鼠的血浆胰岛素水平。不同治疗对空腹胰岛素水平有显着影响(F(3,28)= 7.472,P <0.01)。 Examorelin单独增加血浆胰岛素(P <0.05),而STZ显着降低血浆胰岛素(P <0.001)。 STZ + Examorelin治疗的大鼠显示胰岛素水平升高(P <0.001)。与空腹胰岛素水平相似,喂食胰岛素水平(治疗(3,28)= 61.89,P <0.001)在Examorelin治疗的大鼠中显着增加(P <0.001)并且在STZ治疗的大鼠中减少。此外,Examorelin还改善了STZ诱导的血浆胰岛素水平降低[2]。
细胞实验	将MIN6和α-TC6细胞接种在96孔板中，并在Examorelin（1μM）和STZ（1mM）存在或不存在下培养0.5,1,2,4和6小时。处理后，用新鲜的SFM替换细胞，并使用含有四唑鎓化合物MTS的CellTiter 96 Aqueous One Solution Cell Proliferation Assay定量活细胞数。通过用酶标仪测量490nm处的吸光度来确定细胞活力，并表示为相对于对照细胞的百分比[2]。
动物实验	大鼠[2]实验在雄性Wistar大鼠（180-200g）中进行。在标准的住房条件下（从06:00到18:00点亮，温度22±2℃）将大鼠分开饲养，并随意获得食物和水。将单次腹膜内注射溶解在0.1mol / L柠檬酸盐缓冲液（pH4.5）中的STZ以65mg / kg体重的剂量使用。年龄匹配的对照大鼠接受等体积的载体（柠檬酸钠缓冲液）。 STZ或载体注射后4周，根据治疗方法将大鼠随机分为4组（n = 16 /组）：载体，然后用盐水给予大鼠2周（对照组）;或100μg/ kg Examorelin，持续2周（Examorelin治疗组）; STZ处理的大鼠接着盐水处理2周（STZ处理组）;或100μg/ kg Examorelin，持续2周（STZ + Examorelin治疗组）。每天腹腔注射盐水和Examorelin。
