nAChR[1]

体外研究 侧脑室注射(±)-Epibatidine(0.1μg)显着增加了相互作用间隔(ICI)但不改变压力阈值(PT)或最大排尿压力(MVP),而1μg(±)-Epibatidine增加PT和MVP( P <0.05)和ICI降低。静脉注射低剂量(±)-Epibatidine(0.001-0.1μg)无效;然而,大剂量的(±)-Epibatidine(1μg)显着降低ICI并增加MVP(P <0.05)但不改变PT(P> 0.05)。 (±)-Epibatidine不仅对脑中的nAChR亚型(α4β2)有强效刺激作用,而且对交感神经和副交感神经节(α3β4)以及神经肌肉接头(α1β1γδ)也有强效刺激作用[1]。
动物实验 大鼠[1]在氨基甲酸酯麻醉的(1.2g / kg sc)或清醒的雌性Sprague-Dawley大鼠(250-300g)中研究空隙。在所有实验中,在脑室内和静脉内注射媒介物或(±)-Epibatidine溶液之前,记录对照膀胱测压图(CMG)约2小时。剂量 - 反应曲线通过增加剂量的(±)-Epibatidine [0.001-1μg在1μL脑室内(icv))构建;在200μL静脉注射中0.001-1μg,30分钟至2小时间隔。载体后约30分钟给予(±)-Epibatidine(aCSF,1μLicv;或盐水溶液,200μL,iv)。在一些实验中,通过脑室内途径在(±)-Epibatidine之前10-30分钟注射氯化萘胺(10μg,1μLicv)以阻断激动剂的作用。测量引起排尿(PT),MVP和收缩间隔(ICI;空隙之间的间隔或反射性膀胱收缩)的膀胱内压力,并将其转换为对照值的百分比变化[1]。
