体外研究 谷氨酰胺酶-IN-1(CPD20),一种CB839衍生物,是含有1,3,4-硒二唑的肾型谷氨酰胺酶(KGA)的变构抑制剂,IC50为1 nM。谷氨酰胺酶-IN-1显示出改善的细胞摄取和抗肿瘤活性。谷氨酰胺酶-IN-1的IC 50值分别在A549,H2,Caki-1和HCT116细胞系中分别为17nM,6.78μM,19nM和9nM。谷氨酰胺酶-IN-1比相应的BPTES和CB839具有更好的KGA抑制活性[1]。
体内研究 谷氨酰胺酶-IN-1(10mg/kg,ih)减小了HCT116肿瘤的大小和重量,并且统计分析显示CPD20的肿瘤重量减少40%是统计学显着的。谷氨酰胺酶-IN-1可以在统计学上显着延长携带H22的小鼠的存活[1]。
