描述 FAK-IN-5(化合物8l)是一种FAK信号抑制剂。FAK-IN-5诱导细胞凋亡和自噬[1]。

体外研究 FAK-IN-5(化合物8l)(0-50µM,72小时)对癌细胞具有抗增殖活性[1]。FAK-IN-5(化合物8l)导致细胞分离[1]。FAK-IN-5(化合物8l)(0-25µM,24小时)诱导HCT-116细胞自噬和凋亡【1】。细胞增殖试验【1】细胞系:K562、CEM、G361、MCF-7和HCT-116浓度:0-50µM培养时间:72 h结果:对K562、CEM、G361、MCF-7和HCT-116细胞显示抗增殖活性,IC50值分别为6.3、7.9、6.3、5.5和5.3µM。Western Blot分析【1】细胞系:HCT-116浓度:1.25、2.5、5、7、10、11、17和25µM孵育时间:1、3、5、7和24 h结果:导致FAK、p130Cas和帕西林的去磷酸化呈剂量依赖性。在更早的时间点观察到Y397处FAK的去磷酸化。检测激活的caspase-7片段以及聚ADP核糖聚合酶(PARP)的剂量依赖性切割。
