体外研究 | 肝素是一种有效的抗凝血药物,基于其加速抗凝血酶抑制血液凝固级联中丝氨酸蛋白酶的速率的能力。肝素与含有高正电荷密度的互补结合位点的肽最紧密地相互作用。肝素类似于DNA,因为它们都是高电荷的线性聚合物,其表现为聚电解质。肝素被认为主要通过其与AT III的相互作用起抗凝作用,通过增强AT-III介导的凝血因子(包括凝血酶和因子Xa)的抑制。肝素以三元复合物与AT III和凝血酶结合,使凝血酶抑制的双分子速率恒定增加2000倍。肝素主要位于与免疫应答密切相关的组织肥大细胞颗粒中。肝素与FGF-2和FGFR-1的多种接触稳定FGF-FGFR结合。肝素还与邻近的FGF-FGFR复合物的FGFR-1接触,因此似乎促进FGFR二聚化[1]。 |
