2-氧丙酸钠(丙酮酸钠)是葡萄糖的三碳代谢产物,是糖酵解途径中产生的化合物。2-氧丙酸钠是一种自由基清除剂,可清除活性氧[1][2]。

[体外研究]

在清除过氧化氢的过程中,2-氧丙酸钠(丙酮酸钠)被脱羧为乳酸,因此远离有氧代谢和进一步的活性氧生成。在丙酮酸刺激JNK1活性的条件下,线粒体和胞浆ROS水平均升高。在丙酮酸浓度范围内,观察到多种不同细胞类型的JNK1活性增加[1]。2-氧丙酸钠(丙酮酸钠)是H2O2和O2的有效清除剂▪-, 因此,在体外和体内条件下,2-氧丙酸钠可保护晶状体免受氧化应激和随后的白内障形成,通过竞争性抑制糖羰基和蛋白质-NH2之间的初始反应,2-氧丙酸钠也可保护晶状体蛋白质免受糖基化[2]。

[体内研究]

给新生大鼠一团2-氧丙酸钠(丙酮酸钠；0.1-10 g/kg),1小时后,测定肝提取物中JNK1活性水平。添加2-氧丙酸钠(丙酮酸钠)导致JNK1活性显著增加。对新生大鼠血清2-氧丙酸钠水平的分析表明,丙酮酸的基线水平为0.30mm。在献祭时,在最大剂量的2-氧丙酸钠之后,血清水平上升约六倍,达到1.84 mM[1]。
