柠檬酸三钠是一种天然防腐剂和食品酸味增强剂。柠檬酸三钠诱导G2/M期和S期细胞凋亡和细胞周期阻滞。柠檬酸三钠通过降低抗氧化酶活性导致肝脏氧化损伤。柠檬酸三钠对小鼠造成肾毒性[1][2][3]。

[体外研究]

柠檬酸三钠(0-12.5 mM;24 h)具有剂量依赖性的抗增殖活性[3]。柠檬酸三钠(12.5 mM;72 h)以剂量依赖性方式诱导G2/M期和S期细胞凋亡和细胞周期停滞[3]。柠檬酸三钠(12.5 mM;48 h)增加FAS、BAX、BID、AIF、EndoG、细胞色素c、PARP、GADD153、GRP78和caspase-3、-8、-9的表达,并降低BCL-2和BCL-Xl[3]。细胞活力测定[3]细胞系:HaCaT细胞浓度:0、2.5、5、7.5、10、12.5 mM培养时间:24 h结果:以剂量依赖性方式抑制细胞活力。细胞周期分析[3]细胞株:HaCaT细胞浓度:12.5 mM培养时间:0、12、24、48、72 h结果:诱导细胞凋亡,细胞周期阻滞于G2/M期和S期,呈剂量依赖性。Western Blot分析[3]细胞株:HaCaT细胞浓度:12.5 mM培养时间:12、24、48 h结果:FAS、BAX、BID、AIF、EndoG、细胞色素c、PARP、GADD153、GRP78和caspase-3、-8、-9的表达增加,BCL-2和BCL-Xl的表达降低。

[体内研究]

柠檬酸三钠(120、240和480 mg/kg;i.p.)显著降低小鼠肝脏中GSH-Px活性,并诱导MDA(丙二醛)水平升高[1]。柠檬酸三钠(120、240和480 mg/kg;i.p.)通过以剂量依赖性方式增加小鼠肝细胞中caspase-3活性,诱导细胞凋亡[1]。柠檬酸三钠(120、240和480 mg/kg;i.p.;每周,持续3周)对小鼠造成肾毒性[2]。动物模型:20 g雄性昆明小鼠[2]剂量:120、240、480 mg/kg给药:I.p。;结果:随着柠檬酸剂量的增加,治疗组的T-SOD和GSH-Px活性降低,NOS活性趋于升高,H2O2和MDA含量逐渐降低。
